Dimethyltryptamine (DMT) is made from synthesize from animal and plant which react as a
hallucinogen. The systematic name is N, N- dimethyl-1H-indole-3-ethanamine. In addition, the sigma-1
receptor of cell can distribute with the central nerves system and periphery. There are located in a
plasma membrane, at the endoplasmic reticulum that adjust calcium [Ca+] signaling through the IP3
receptor. The sigma1 receptor are a chaperone protein which are cloned human and rodent, no
homology in other mammalian protein. Furthermore, the alkylamine core can also found in N, Ndimethyltryptamine. It is bound to sigma-1 receptor and inhibit voltage gate sodium ion channels. DMT
is important reaction with the sigma-1-receptor.
The purpose is to extract dimethyltryptamine and examine that how is it work with cancer. The first
synthesize from amino acid is called tryptophan. The systematic name is (2s)-2-amino- 3-(1H- indol-3-yl)
or propionic acid. After the synthesize tryptophan, it will be a tryptamine. Dimethyltryptamine is the last
step of synthesize from tryptamine.
Procedure:
Tryptophan to Tryptamine
Prepared tryptophan 454g suspended in tertralin 1150ml and acetone 129g
The mixture was heated for 12 hours with vigorous stirring until no carbon dioxide
was evolved
Then the reaction mixture became clear, the solvent was remove under vacuum.
The residuum was distilled under reduce pressure to give a yellow crystalline solid.
Tryptamine to DMT
Prepare tryptamine 1.12 g. 7 mmol , sodium Cyan borohydride 0.88g 14 mmol and
glacial acetic Acid 2ml 35mmol. Then dissolve all of these in Methanol 110 ml at 0C
Added dropwise over 20 minute and solution Formaldehyde 37% 1.4 ml, 18.5 mmol.
(Allow to stir for 20 minute at 0C and 2.5h at room temperature).
The methanol become vapor under reduce pressure.
Add 80ml 25% of aqueous of potassium carbonate and the solution extracted with
2125ml EtOAc. The extract wash with 240ml brine.
Dried over MgSO4 and the solvent become vapor under reduce pressure to give an
amber oil which was purified by flash chromatography on 30g silica gel.
Using gradient of EtOAc, MeOH to give an oil which was crystallized from boiling
hexane to give N, N-Dimethyltryptamine as colorless waxy crystals, weighing 0.9g
(69%)
