Description
BoltonSelectiveEnrichmentBrothisintendedforthepreenrichmentofCampylobacterinfoodsamples.
CampylobacterareGramnegative,spirallyshapedmicroaerophilicorganismswhichmaybepresentin
rawmilk,untreatedwater,improperlyhandledfoodandundercookedmeats,poultryandshellfish.
Humanconsumptionoftheseorganismscanresultinarangeofclinicalillnessesfromtransient
asymptomaticcolonisationtoseveredysentery.ThesymptomsofCampylobacterenteritisinclude
diarrhoea,stomachpain,nausea,fever,headacheandmusclepain.Complicationsofinfectionby
Campylobacterjejunimayincludeunnecessaryappendectomiesasaresultofabdominalpain,reactive
arthritisorGuillianBarrsyndrome1.Campylobacterinfectionisrecognisedasoneofthemostcommon
causesofbacterialgastroenteritisinhumans,andtheminimuminfectivedosemaybeaslowas500800
cells1.
SinceawarenessoftheapparentroleofCampylobacterinhumandiseasewasheightenedbySkirrowin
19772,agreatnumberofculturemediahaveevolvedinresponsetotheneedtooptimiseperformance.
Therewasearlyrecognitionoftheneedforenrichmentculturewhenexaminingfoodsamplesto
overcomethedamagingeffectsthatfoodprocessingandpreservationtechniquescanhaveon
Campylobactercells.Useoflowerincubationtemperaturesintheearlystagesofenrichmentisnow
widelyestablishedasanaidtocellrecovery3.ThisprinciplewasemployedbyBoltoninthedevelopment
ofhisenrichmentbroth4.
Campylobactercanbeinjuredbyfoodprocessingandpreservationprocedures3.Thismakesthem
susceptibletoselectiveagentswhicharetoleratedbyundamagedcells.Falsenegativeresultsare
avoidedthroughuseofrecoverymediumsuchasBoltonSelectiveEnrichmentBrothwhichincreasesthe
numberofcellsavailableforculture,firstbyresuscitatinginjuredorganismsandthenencouragingthem

Campylobacterspp.VancomycinactiveagainstGrampositives.Cefoperazonepredominantlyactive
againstGramnegatives.TrimethoprimactiveagainstawidevarietyofGramnegativeandGram
positiveorganisms.Cycloheximideactiveagainstyeasts.
Technique
Onemethodofuseisasfollows:
Place25goffoodsamplein225mlBoltonSelectiveEnrichmentBroth(preparedasdescribedabove)
andhomogenisethemixtureusingaStomacher(orsimilardevice).BoltonSelectiveEnrichmentBroth
doesnotrequireincubationinamicroaerobicenvironment,butmustbeusedinscrewtoppedcontainers
whicharefilledtowithin20mmofthetop4.Incubatefor4hoursat37C,followedbyfurtherincubation
at42C.Thebrothcanbesubculturedafter24hoursand48hoursontoeitherModifiedCCDA
(CM0739+SR0155)orPrestonAgar(CM0689+SR0117+SR0048)4.
