AEI603.37 1
Objectives

On completion of this topic you would be

able to know about Liquid Chromatography

• Introduction

• Block diagram

AEI603.37 2
Recap

In last class we have studied about gas

chromatograph
• Introduction

• Principle

• Operation

• Applications

• Detectors

AEI603.37 3
Introduction

• Chromatography is a physical method of separation of

the components of a mixture by distribution between two

phases

• One is a stationary phase of large area

• Other is a mobile phase which passes along the

stationary phase

AEI603.37 4
Definition

• If the mobile phase is liquid then the chromatography is

called “ Liquid Chromatography”

AEI603.37 5
High Pressure Liquid Chromatography
(HPLC)

• It can be considered as the oldest of all chromatographic

process

• High Pressure Liquid Chromatography (HPLC) is similar

to gas chromatography

• But the process is carried through in liquid form by a

solvent

AEI603.37 6
High Pressure Liquid Chromatography
(HPLC) …….

• The substance was not be vaporized

• HPLC can be used where the substance can not be

analyzed using gas chromatography

• The use of pressure increases the linear velocity

AEI603.37 7
High Pressure Liquid Chromatography
(HPLC) …..

• It gives the components to less time to diffuse within the

column

• Improves the resolution of the chromatogram

• It introduces a refinement called the Gradient elution

• i.e. variation of the mobile phase during the analysis
